Isolation and genotyping of Nile tilapia MHC IA gene

Abstract
Aim: This study was conducted to isolate the major histocompatibility complex (MHC) class IA gene from Nile tilapia and to compare between different genotypes of this gene following infection with Aeromonas hydrophila. Materials and methods: MHC IA gene was amplified by PCR using genomic DNA extracted from whole blood samples of control (unchallenged) and diseased (challenged by A. hydrophila) fish. Genotyping was performed by single strand conformational polymorphisms (SSCP) technique and genotypic and allelic frequencies were then determined. Results: PCR revealed amplification of MHC IA with a size of 568 bp. SSCP showed three banding patterns (GG, GC, and CC). The frequency of GG was higher in control fish than CC, however in diseased fish, CC genotype was higher than GG. Consequently, the frequency of C allele was significantly higher in diseased fish than control fish. Moreover, the three genotypes were in Hardy–Weinberg disequilibrium (HWE) because χ2 value was 0.13 (P > 0.05). Conclusion: These information indicate that C may be the susceptibility allele to A. hydrophila infection in Nile tilapia. When compared to the genotypes for the MHC IA gene, brood stock can only be chosen with the resistant alleles in diseased and controlled fish.
