Effect of myostatin inhibitor (myostatin-propeptide) on expression of myogenic genes in chick embryo

Abstract
Aim: This study aimed to investigate the underlying mechanism by which myostatin (MSTN) inhibitor MSTN-propeptide (MSTNP) could regulate the expression of some muscle growth-related genes, Pax3, Pax7, MyoD and MyoG in chick embryo.
Materials and methods: The pre-somitic mesoderm (PSM) of HH7 Fayoumi chick embryos (n = 100) were injected with either empty retroviral vector (RCAS) in the control group, or RCAS containing MSTNP gene (MSTNP group). Real time PCR (qPCR) was used to monitor changes in genes expression.
Results: Chick embryos injected with MSTNP showed significant upregulated expression of muscle growth-related genes, Pax3, Pax7, MyoD and MyoG, with highest expression for Pax7, followed by Pax3, then MyoG and finally MyoDin chick embryos as compared to the control group.
Conclusions: From the obtained results, MSTN inhibition is accompanied by induction of Pax3, Pax7, MyoD and MyoG genes upregulation in chick embryos.
