Immunohistochemical and molecular changes in the ovary of Egyptian buffalo during follicular and luteal phases

Abstract
Background: Fertility related growth factors such as BMP15, GDF9, IGF1, IGF2, IGF1R and FSHR have been found to play a crucial role in oocyte differentiation during oogenesis and folliculogenesis. However, the detailed expression profile of these factors and their encoded proteins was not determined yet in Egyptian buffaloes during different phases of estrus cycle. Aim: The present study was conducted to investigate immunohistochemical and molecular changes occur in ovary of Egyptian buffalo during the follicular and luteal phases of estrus cycle. Materials and methods: A total number of fifty ovaries were collected and divided into three groups (non-cycling, follicular and luteal) depending on structures appeared on the ovarian surface. Twenty ovaries were fixed in neutral formaldehyde (for immunohistochemistry) and the remaining were processed for RNA extraction (for gene expression using real time PCR). Results: The obtained results showed a significant up-regulation in the expression BMP15, GDF9, IGF1, IGF1R, IGF2 and FSHR genes in ovarian tissues of the cycling animals during follicular and luteal phases, except for BMP15, GDF9 and FSHR in the luteal phase which showed significantly lower expression, as compared to the non-cycling animals. The follicular phase exhibited significantly higher gene expression and positive FSHR immunostaining than the luteal phase. Conclusion: The results of this study suggest that BMP15, GDF9, IGFI, IGFIR, IGFII and FSHR may play a key role in follicular development, differentiation and also in luteal activity in Egyptian buffalo ovary.
