Costus speciosus extracts relieve lead acetate-induced neurotoxicity in rats

Abstract
Several Costus speciosus extracts have the ability to reduce the adverse effect of some toxicants, but ability of C. speciosus to relieve lead acetate (PbAc)-induced toxicity has not fully investigated yet. Hence, this study was conducted to evaluate the effect of aqueous, petroleum ether, and methanolic extracts of C. speciosus rhizome on PbAc-induced neuro-toxicity in rats. Male albino rats were divided into 5 groups (n = 30/group) and each group was subdivided into 3 subgroups (n = 10/subgroup) dependant on duration (10, 20, and 30 days): group I (control), group II received PbAc (50 mg/kg body weight), and groups III-V co-administrated aqueous, petroleum ether, and methanolic extracts of C. speciosus, respectively with PbAc. Brains were collected and weighted then immediately kept in liquid nitrogen and used for determination the levels of acetylcholine (ACh), dopamine (DA) and serotonin or 5-hydroxy tryptamine (5-HT) and the expression of acetylcholinesterase (AchE), dopamine receptor 2 (Drd2) and tryptophan hydroxylase (TPH1) genes. All extracts had relieved the neuro-toxic effect of PbAc as revealed by inhibition of ACh and AchE and induction of DA and 5-HT and their target genes Drd2 and TPH1. Long time treatment (for 30 day) with methanolic extract showed the best ameliorative effect on PbAc neurotoxicity. It could be concluded that C. speciosus extracts possess improvement potentials on PbAc-induced neuro-toxicities through, at least in part, induction of dopamine and serotonin that further activate their target genes such as Drd2 and TPH1 to overcome PbAc-induced neurotoxic stress. Therefore these extracts, especially the methanolic one, could be useful ameliorative agents against the adverse effect of PbAc on nervous tissues.
